However, mechanisms controlling the differentiation of NPCs are not well understood. Using calcium (Ca2+) imaging, we examined intracellular Ca2+ fluctuation that is frequently observed during embryonic development. We found that the onset of the intermittent intracellular Ca2+ transients in NPC intracellular Ca2+ fluctuation is correlated to NPC differentiation toward neurons. Furthermore, the intracellular Ca2+ fluctuation in NPC was mediated by the T-type Ca2+ channel and blockage of this Ca2+ channel inhibited neuronal differentiation.
