| 内容記述 | 微小管結合タンパク質「タウ」の凝集体が細胞内に蓄積する神経変性疾患をタウオパチーという。本研究では、タウの凝集メカニズムを明らかにするため、マウス脳の微小管安定性およびタウー微小管結合状態の解析を行った。結果、非タウオパチー脳は、タウオパチー脳よりも遊離型タウが少ないことが明らかになった。さらに、生理的な遊離型タウと結合するタンパク質が複数同定された。以上より、遊離型タウが凝集に関与し、遊離型タウ結合タンパク質がタウの微小管結合領域の露出と凝集を抑制していることが示唆された。 | |||||||||||||

| 内容記述 | Tauopathies are neurodegenerative diseases defined by the accumulation of misfolded, insolubule microtubule-associated peotein "tau" aggregates. In this study, to clarify the mechanism of tau aggregation, the microtubule stability and tau-microtubule binding state in mouse brains were analyzed. As a result, non-tauopathy brains had less free tau than tauopathy brains. Furthermore, several proteins that bind to physiological free tau was identified. These results suggested that free tau is involved in aggregation, and that free tau-binding proteins suppress the exposure of the microtubule-binding domain of tau and its aggregation. | |||||||||||||
